摘要: Nine different maleimide compounds were evaluated for the conjugation of Fab' to horseradish peroxidase through thiol groups in hinge. The succinimidyl maleimidoacetate (I), 4-maleimidobutyrate (II), 6-maleimidohexanoate (III), 4-(N-maleimidomethyl)cyclohexane-1-carboxylate (IV), m-maleimidobenzoate (V), 4-(p-maleimidophenyl)butyrate (VI), sulfosuccinimidyl (VII), (VIII), and (IX). Maleimide I-IV VII fairly stable at pH 7.0 30 degrees C, while those other significantly decomposed. I-III VII-IX sufficiently soluble reaction mixture introduction groups, others more or less precipitated during reaction. II III most effective gave highest recovery conjugate, which reached 80%. From these results, judged be useful, IV useful.
