摘要: The alkaline comet assay is a sensitive test for the detection of variety DNA lesions. However, crosslinks are not readily detected under standard conditions. Recently, modifications have been introduced measuring by determining reduction induced migration. We used to comparatively investigate in V79 cells effect three different crosslinkers: formaldehyde (FA), which predominantly induces DNA-protein crosslinks, cisplatin (DDP), mainly produces DNA-DNA-intrastrand and mitomycin C (MMC), leads DNA-DNA-interstrand crosslinks. In assay, only MMC slight increase migration at high toxic concentrations. FA DDP did induce any conditions used. modified all crosslinkers led clear gamma-ray-induced This was seen case parallel induction cytotoxicity SCE, while cytotoxicity, SCE HPRT gene mutations occurred much lower concentrations than effects assay. DNA-DNA caused cytotoxic Our results indicate that protocol suggest well suited evaluation crosslinkers. Furthermore, relationship between crosslinking genotoxicity seems be very types substances.
