Wheat germ systems for cell‐free protein expression

摘要: Cell-free protein expression plays an important role in biochemical research. However, only recent developments led to new methods rapidly synthesize preparative amounts of that make cell-free attractive alternative cell-based methods. In particular the wheat germ system provides highest translation efficiency among eukaryotic approaches and has a very high success rate for soluble proteins good quality. As open vitro method, is preferable choice many applications research including options labeling difficult-to-express like membrane multiple complexes. Here I describe systems give examples how they have been used genome-wide studies, preparation labeled structural genomics mass spectroscopy, automated synthesis, screening enzymatic activities. Future directions use are discussed.
