The effects of acute and preventive migraine therapies in a mouse model of chronic migraine

摘要: Background The development of novel migraine therapies has been slow, in part because the small number clinically relevant animal models. We have recently developed a new mouse model chronic using intermittent nitroglycerin, known human trigger. objective this study was to validate by testing and potential migraine-preventive treatments. Methods Migraine were administered male female mice for 11 days. On day 3, tested with nitroglycerin every second nine Basal nitroglycerin-evoked mechanical hypersensitivity evaluated von Frey filaments. Results Chronic produced acute hyperalgesia each administration, progressive sustained basal hypersensitivity. established preventive therapy propranolol effectively blocked nitroglycerin-induced hyperalgesia, while valproate had no effect. Potential also tested: Amiloride inhibited hyperalgesia; memantine ineffective. sumatriptan, which did not alter but instead resulted similar that observed following administration. Conclusions This establishes as an additional screening tool test therapies.
