The Number of Mitochondrial Deoxyribonucleic Acid Genomes in Mouse L and Human HeLa Cells

摘要: Abstract Mitochondrial DNA can be isotopically labeled to the virtual exclusion of nuclear labeling in cell lines lacking major soluble thymidine kinase (EC 2.7.1.21) but retaining a mitochondrial kinase. This characteristic provides means for selective assay during isolation and determination cellular content DNA. The simplest three procedures compared this study is shown yield approximately two-thirds total tissue culture cells examined. Mouse L containing predominantly 107 dalton closed circular species have 1100 ± 250 molecules per cell. A second line mouse which has molecular weight 2 x contains 900 HeLa at least four times mass volume as cells.
