Sequence characterization of the adenovirus 40 fiber gene

摘要: Abstract The fiber gene of human adenovirus type 40 has been characterized. 6.1-kbp EcoRl fragment C the Ad40 genome, from map units 74 through 92, was cloned and right-most 2.8 kbp 84 sequenced. By analogy with Ad2, this region would be expected to contain specifying polypeptide. Sequencing revealed an open reading frame 1641 bases on r-strand, first 53 which had marked homology corresponding L5 (fiber) regions Ad2 (77.2%), Ad5 (75.0%), Ad3 (64.3%). In addition, base positions 1114 1146 85% 1198 1230 gene. predicted polypeptide sequence 547 amino acids showed Ad5, polypeptides in two regions, 55 N-terminus 372 382. Analysis hydrophobic acid a repeating pattern approximately 15 residues between 42 374, 21 repeats. thus fits model Green et al. ((1983), EMBO J. 2, 1357–1365) for structure fiber, but is 35 shorter than polypeptide, one less 15-residue repeat shaft region. According model, highest AD40 those correspond tail knob. results analysis are agreement previously published EM data length subgroup F adenoviruses.
