Submerged fermentation of wheat bran by Aspergillus flavus for production and characterization of carboxy methyl cellulase

摘要: Abstract Objective To elucidate the capabilities of carboxy methyl cellulase (CMCase) production with selective species Aspergillus flavus (A. flavus) . Methods The expression in A. was evaluated under different environmental conditions using submerged fermentation (SmF) on agricultural byproducts. Results produced high levels CMCase optimized culture 3 rd day incubation at an optimum pH 6.0, temperature 30 °C, inoculum size 4% Czapek Dox wheat bran as a substrate by SmF. Enhanced occurred addition and 1% peptone nutritional factors. enzyme hydrolyzed exhibiting V max value 40 U/mg K m 0.5 mg/mL 7. optimally activated 50 °C. Except Mg 2+ , all other metal ions such Zn Cu Na + Ca were found to be activators for activity. Conclusion kinetic parameters showed thermo stability, stability which reflects potential commercial significance enzyme.
