摘要: Biochemical and genetic studies support the view that majority of DNA double-strand breaks induced in genome higher eukaryotes by ionizing radiation are removed two pathways nonhomologous end joining (NHEJ) termed D-NHEJ B-NHEJ. Whereas depends on activities DNA-dependent protein kinase ligase IV/XRCC4, components B-NHEJ have not been identified. Using extract fractionation, we show activity extracts HeLa cells derives from III. III fractionates through columns with maximum its depletion causes loss can be recovered addition purified enzyme. The same fractionation protocols provide evidence for an additional factor strongly enhancing shifting product spectrum circles to multimers. An vivo plasmid assay shows IV–deficient mouse embryo fibroblasts retain significant reduced up 80% knocking down using RNA interference. These vitro observations identify as a candidate component point factors contributing NHEJ efficiency.
