Co-carcinogenic effects of vitamin E in prostate.

摘要: A large number of basic researches and observational studies suggested the cancer preventive activity vitamin E, but large-scale human intervention trials have yielded disappointing results actually showed a higher incidence prostate although mechanisms underlying increased risk remain largely unknown. Here we show through in vitro vivo that E produces marked inductive effect on carcinogen-bioactivating enzymes pro-oxidant status promoting both DNA damage cell transformation frequency. First, found epithelial RWPE-1 line has remarkable ability to upregulate expression various phase-I activating cytochrome P450 (CYP) enzymes, including activators polycyclic aromatic hydrocarbons (PAHs), giving rise supraphysiological levels reactive oxygen species. Furthermore, our rat model confirmed powerful booster CYP associated with generation oxidative stress, thereby favoring lipid-derived electrophile spread covalently modifies proteins. We not only causes also promotes frequency induced by PAH-prototype benzo[a]pyrene. Our findings might explain why dietary supplementation increases among healthy men.
