Chemical inactivation of soybean trypsin inhibitors

摘要: Sodium metabisulfite and glutaraldehyde were used alone in combination to inactive Kunitz trypsin inhibitor (TI) model systems TI lyophilized alkaline soy meal extract. Reaction of (0.1 3.0%, based on total volume the reaction mixture) with (3 mg/ml buffer) at 25 C resulted 60–75% reduction activity. Treatment extract containing 0.08 mg TI/mg a concentration 10 sample/ml buffer under similar conditions reduced activity only 40%, however. A temperature 75 had little additional effect inactivation. On other hand, sodium (0.05 1.0 mM) inactivated >96% within 1 hr 75–94% The 0.6 mM 0.5% >99% TI, up 3% 88% vs 91% inactivation alone. Thus, plus better than either one alone, while bisulfite best When reactions performed neutral pH, protein solubility 80% or was retained.
