摘要: A variant of the whole-cell patch clamp technique is described which allows measurement ionic currents in small cells while minimizing cell dialysis with pipette solution. The involves application negative pressure to inside (< 1 µm) tip diameter pipettes placed on surface achieve high resistance seals and membrane rupture. has been used successfully a variety different types study carried by Ca K, generated exchange carriers as well electrical coupling between cells. Overall, seems suited for cells, provides an alternative conventional clamping techniques necessitate intracellular dialysis.
