摘要: LEF-1 is a transcription factor that participates in the regulation of T-cell receptor alpha (TCR alpha) enhancer by facilitating assembly multiple proteins into higher order nucleoprotein complex. The function dependent, part, on HMG domain induces sharp bend DNA helix, and an activation stimulates only specific context other enhancer-binding proteins. With aim gaining insight context-dependent domains, we cloned ALY, novel LEF-1-interacting protein. ALY ubiquitously expressed, nuclear protein specifically associates with domains AML-1 (CBF alpha2, PEBP2 alpha(B), which another component TCR In addition, can increase binding both AML Overexpression activity complex reconstituted transfected nonlymphoid HeLa cells, whereas down-regulation anti-sense oligonucleotides virtually eliminates T cells. Similar to LEF-1, stimulate but apparently not when tethered through heterologous DNA-binding domain. We propose mediates transcriptional functional collaboration
