摘要: Commitment of cells to apoptosis is governed largely by protein-protein interactions between members the Bcl-2 protein family. Its three sub-families have distinct roles: BH3-only proteins trigger binding via their BH3 domain pro-survival relatives, while pro-apoptotic Bax and Bak an essential downstream role involving disruption organellar membranes induction caspase activation. The act as damage sensors, held inert until activation stress signals. Once activated, they were thought bind promiscuously targets but unexpected selectivity has recently emerged from analysis interactions. Some also Bak. Whether are activated directly these proteins, or indirectly a consequence neutralizing subject intense debate. Regardless this, detailed understanding family members, which often selective, notable implications for designing anti-cancer drugs target
