5-Azacitidine Induces Cell Death in a Tissue Culture of Brachypodium distachyon

摘要: Morphological and histological observations revealed that, at a concentration of 50 µM, 5-azacitidine (5-azaC) totally inhibited the induction embryogenic masses (EM), while cultivation explants (zygotic embryos; ZEs) in presence 5 µM 5-azaC led to formation callus with EM 10% cases. Transmission electron microscopy (TEM) analyzes morphological ultrastructural features that are typical for vacuolar type cell death cells were treated. A TUNEL assay confirmed DNA double-strand breaks had been treated both concentrations. Analysis gene expression selected markers demonstrated reduced metacaspase, protein executer 1 (EX1), thioredoxin (TRX) compared control culture. The strongest increase activity was characteristic glutathione S-transferase (GST). Our studies also included an analysis distribution some arabinogalactan proteins (AGPs) extensin epitopes, which can be used as undergoing Brachypodium distachyon tissue
