Hematologic reference intervals for koi (Cyprinus carpio), including blood cell morphology, cytochemistry, and ultrastructure.

摘要: Background Hematologic data are used routinely in the health care of humans and domestic mammals. Similar for fish largely fragmentary or have not been collected. Objectives The primary purpose this study was to determine hematologic reference intervals koi, an ornamental strain common carp (Cyprinus carpio). Secondarily, morphology, cytochemical reactions, ultrastructure koi blood cells were characterized. Methods A CBC performed manually on heparin-anticoagulated specimens using Natt Herrick's diluent a Neubauer-ruled hemacytometer. Leukocyte differential counts done Wright-Leishman- Diff-Quik-stained smears. Cytochemical reactions leukocytes determined commercial kits. Transmission electron microscopy characterize ultrastructural features cells. Results established healthy PCV (30-34%), hemoglobin concentration (6.3-7.6 g/dL), RBC count (1.7-1.9 X 10(6)/ microL), WBC (19.8-28.1 10(3)/ indices, leukocyte counts. Lymphocytes predominant (accounting up 80% all leukocytes), whereas eosinophils rare. Basophils positive with PAS staining. Naphthol AS-D chloroacetate esterase activity observed only eosinophils. alpha-Naphthyl butyrate beta-glucuronidase activities monocytes. Some lymphocytes reactive alpha-naphthyl acid phosphatase activity. Ultrastructurally, leukocytes, erythrocytes, thrombocytes identified basis cytoplasmic organelles granule appearance. Conclusion knowledge characteristics will help standardize studies species.
