摘要: To locate genes predisposing to insulin-dependent diabetes mellitus (IDDM), an autoimmune disorder resulting from destruction of the insulin-producing pancreatic cells, we are testing linkage IDDM susceptibility polymorphic markers across genome using families with two or more children. A new locus (IDDM11) has been localized chromosome 14q24.3-q31 by detection significant microsatellite D14S67, both maximum likelihood methods (LOD{sub max} = 4.0 at {theta} 0.20) and affected sib pair (ASP) (P 1 x 10{sup -5}). This represents strongest reported evidence for any outside HLA region. The subset in which children did not show increased sharing (HLA {le}50%) provided most support D14S67 max}4.6 0.12;ASP P < 5 -6}). There was heterogeneity between HLA-defined subsets 0.009), suggesting that IDDM11 may be important lacking strong region predisposition. 52 refs., 2 figs., 3 tabs.
