摘要: Despite exceptional efficacy and safety, fear of muscle toxicity remains a major reason statins are underutilized. Evidence suggests that statin may be mediated by abnormalities in lipid metabolism. To test the hypothesis myotubes from patients intolerant lipid-lowering therapies have abnormal fatty acid oxidation (FAO) responses we compared 11 subjects with intolerance (Intolerant) seven statin-naive volunteers undergoing knee arthroplasty (Comparator). Gross pathology was graded skeletal cell cultures were produced each subject. FAO assessed following treatment increasing concentrations. There no difference biopsy myopathy scores between groups. Basal octanoate greater Intolerant than Comparator (P = 0.03). Lovastatin-stimulated palmitate tended to for Control subjects’ 0.07 5 μM P 0.06 20 lovastatin).
