摘要: During interphase, chromatin hosts fundamental cellular processes, such as gene expression, DNA replication and damage repair. To analyze on a proteomic scale, we have developed enrichment for proteomics (ChEP), which is simple biochemical procedure that enriches interphase in all its complexity. It enables researchers to take 'snapshot' of isolate identify even transiently bound factors. In ChEP, cells are fixed with formaldehyde; subsequently, together cross-linked proteins isolated by centrifugation under denaturing conditions. This approach the analysis global composition changes, contrast existing procedures, either focus specific loci (e.g., affinity purification) or limited specificity, pellet (i.e., insoluble nuclear material). ChEP takes half day complete requires no specialized laboratory skills equipment. characterization response drug treatment physiological processes. Beyond proteomics, may preclear immunoprecipitation (ChIP) analyses.
