Do antiosteoporotic drugs improve bone regeneration in vivo

摘要: Treatment of complex fractures in the elderly is a challenge for operative reconstruction due to degraded bone structure. Early peri-operative anabolic treatment could improve new formation, avoid implant loosening and accelerate fracture healing. To compare osteoanabolic potential different drugs after distraction osteogenesis, 168 female Sprague–Dawley rats underwent lengthening right femur using monolateral external fixator. Animals were randomly divided into six groups: vehicle-injected group, PTH(1-34), raloxifen, strontium ranelate, alendronate simvastatin. Histomorphometry, CT-scanning, DEXA- biomechanical analysis performed evaluate callus volume, mineralisation strength. Expression metabolic mediators differentiation indicators distracted intact examined by RT-PCR western blot. Histological showed significant increase mass with raloxifen ranelate (p = 0.02). Raloxifen increased mineral content (BMC) whole significantly (p = 0.007). Callus volume was larger simvastatin groups (p = 0.001) compared control. Ultimate load formed PTH(1-34) groups. It seems that have stronger effect on where repair response activated. Strontium demonstrates similar effects PTH regarding formation but shows low values This study suggests and/or osteoporotic might be useful as stimulator shorten consolidation time humans.
