摘要: The Class II Transactivator (CIITA) is the master regulator of Major Histocompatibility (MHC II) genes. Transcription CIITA through IFN-γ inducible promoter IV (CIITA pIV) during activation characterized by a decrease in trimethylation histone H3 lysine 27 (H3K27me3), catalyzed methyltransferase Enhancer Zeste Homolog 2 (EZH2). While EZH2 known catalytic subunit Polycomb Repressive Complex (PRC2) and present at inactive pIV, mechanism PRC2 recruitment to mammalian promoters remains unknown. Here we identify two DNA-binding proteins, which interact with regulate pIV. We demonstrate Yin Yang 1 (YY1) Jumonji domain containing protein (JARID2) are binding partners along cells. Upon stimulation, YY1 dissociates from pIV while JARID2 increases, suggesting novel roles for these proteins regulating expression Knockdown yields decreased H3K27me3 important knockdown also results significantly elevated levels mRNA upon stimulation. This study first epigenetic regulation PRC2. Our observations indicate importance silencing, provide YY1-JARID2-PRC2 regulatory complex as possible explanation differential versus permanently silenced
