摘要: Effects of 4-(p-chlorobenzyl)-2-(hexahydro-1-methyl-1H-azepin-4-yl)-(2H)-phthalazinone hydrochloride (azelastine hydrochloride), a new antiallergic drug, on the gastrointestinal tract were experimentally studied in comparison with diphenhydramine and clemastine. In isolated guinea pig ileum, dose-response curve for histamine was shifted to right by 10(-8) mol/l diphenhydramine, clemastine or azelastine same degree. Clemastine reduced maximum response curve, while caused parallel shift. addition potent antihistamine action, at higher dose antagonized serotonin action markedly inhibited contractile responses intestinal preparations acetylcholine bradykinin. Azelastine (50 mg/kg intraduodenally) gastric secretion pylorus ligated rats. (5 i.v.), (1 i.v.) mg/lg depressed motility conscious Biliary pancreatic secretions anesthetized rats not affected 1 i.v.
