摘要: To appreciate the functional diversity of communities soil eukaryotic micro-organisms we evaluated an experimental approach based on construction and screening a cDNA library using polyadenylated mRNA extracted from forest soil. Such contains genes that are expressed by each different organisms forming community represents its metatranscriptome. The contributed to this was sequencing portion 18S rDNA gene amplified either DNA or reverse-transcribed RNA. More than 70% sequences were fungi unicellular eukaryotes (protists) while other most represented group metazoa. Calculation richness estimators suggested more 180 species could be present in samples studied. Sequencing 119 identified with no homologues databases (32%) coding proteins involved biochemical cellular processes. Surprisingly, taxonomic distribution did not coincide, marked under-representation protists among cDNA. Specific such environmental isolated expression heterologous microbial host, Saccharomyces cerevisiae. This is illustrated complementation histidine auxotrophic yeast mutant two originating possibly ascomycete basidiomycete fungal species. Study metatranscriptome has potential uncover adaptations whole local conditions. It also gives access abundant source biotechnological interest.
