摘要: Pellets are one of multiparticulate pharmaceutical forms and can offer numerous technical biopharmaceutical advantages compared with single dose unit formulations, e.g. tablets capsules. This study aimed at formulation controlled-release pellets doxazosin mesylate (DM), a widely used treatment for antihypertensive benign prostatic hyperplasia. DM was loaded onto microcrystalline cellulose CELLETS® using hydroalcoholic solution alcoholic suspension layering techniques to achieve minimum drug load 4 mg DM/g pellets. DM-layered CELLETS were coated by Aquacoat dispersion (ready-made ethylcellulose dispersion) coating pan technique as simple utilized in industry. Controlled-release showed release profile comparable the commercial product Cardura® XL Tablet. Also, mechanism from mathematically modeled imaged scanning electron microscopy elucidate mechanisms prepared pellet formulations. Accelerated stability studies performed under stress conditions 40 °C, 75 % RH 3 months. In conclusion, preparation is feasible conventional technology.
