摘要: A multi-residues method using pressurized liquid extraction (PLE) and chromatography combined with mass spectrometry (LC-MS/MS) has been developed for determination of eight glucocorticoids (prednisone, prednisolone, hydrocortisone, methylprednisolone, dexamethasone, betamethasone, beclomethasone, fludrocortisone) in muscle swine, cattle, sheep. Parameters affecting PLE including solvent, temperature, pressure cycles were optimized. The optimized employed 11 ml cells, hexane-ethyl acetate (50:50, v/v) as 1500 psi 50°C temperature. samples detected by LC-ESI-MS/MS negative mode selected reaction monitoring (SRM) mode. recovery spiked at levels 0.5-6 μg kg(-1) ranged from 70.1% to 103.1%; the between-day relative standard deviations no more than 9.6%. limits quantification 0.5-2 muscle. results demonstrated that is simple, fast, robust, suitable identification residues foods animal origin.
