Genetic and Antigenic Diversity of the Surface Protective Antigen Proteins of Erysipelothrix rhusiopathiae

摘要: The surface protective antigen (Spa) protein of Erysipelothrix rhusiopathiae has been shown to be highly immunogenic and is a potential candidate for new vaccine against erysipelas. In this study, we cloned sequenced spa genes from all E. serovar reference strains as well 18 strain which was not classified any species in the genus Erysipelothrix. Sequence analysis revealed that Spa proteins could into three molecular species, including SpaA, previously found serovars 1a 2, newly designated SpaB SpaC proteins. SpaA produced by 1a, 1b, 5, 8, 9, 12, 15, 16, 17, N, 4, 6, 11, 19, 21, only 18. amino acid sequence similarity high among members each type (96 99%) but low between different types (∼60%). greatest diversity N-terminal half molecule (50 57% similarity), involved immunoprotection. Coinciding with this, immunoblot rabbit antisera specific reacted strongly homologous weakly heterologous A mouse cross-protection study showed recombinant (rSpa) elicited complete protection challenge level varied depending on rSpa used immunization. Our first demonstrate antigenic indicate rSpaC may most promising use component because its broad cross-protectiveness.
