UP-REGULATION OF FAS AND THE COSTIMULATORY MOLECULES B7-1 AND B7-2 ON PERIPHERAL LYMPHOCYTES IN AUTOIMMUNE B6/GLD MICE

摘要: C57BL/6-gld/gld (B6/gld) mice have a point mutation in the gene for Fas ligand (FasL) resulting nonfunctional FasL protein. We hypothesized that lack of normal Fas/FasL interactions these might result abnormalities expression. Thus, we compared spleen cells from B6/gld and B6 control mice. While consisted two main populations, Fashigh (high expression) Faslow (low expression), nearly all were Fashigh. Two-color immunofluorescence revealed populations Thy-1.2+ (T cells) IgM+ (B cells), respectively, whereas both T B Fashigh, indicating expression is increased on spleen. This phenomenon was age related restricted to peripheral lymphocytes. In addition Fas, splenic showed costimulatory molecule B7-2, while B7-1 up-regulated vitro, an increase susceptibility apoptosis mediated by soluble anti-Fas Ab. These results suggest some lymphocytes are primed undergo Fas-mediated apoptosis, but unable do so due absence functional FasL. Further study such abnormal may elucidate nature autoimmunity
