摘要: The site of S-phase DNA synthesis has been the subject recurring controversy. All recent evidence supporting a fixed to some nuclear sub-structure is derived from studies in which cells or nuclei have extracted hypertonic salt concentrations. controversy centres on whether resulting matrices cages counterparts vivo are simply artefacts. Using isotonic conditions throughout isolation and analytic procedures we now reinvestigated replication. Cells encapsulated agarose microbeads lysed leave nevertheless completely accessible enzymes. After incubation with endonucleases, most chromatin can be electroeluted beads: however, nascent active polymerase remain entrapped. Since particles containing size 125 kbp electroelute, conclude that polymerizing complex attached nucleoskeleton too large escape. We also studied various artefacts induced by departure conditions. Perhaps surprisingly, hypotonic used during conventional significant source artefact.
