摘要: Endothelial cell phenotypes are differentially regulated between different sites of the vascular tree. We tested hypothesis that endocan, a novel soluble dermatan sulfate proteoglycan, is expressed in intact endothelium and site-specific expression mediated by signals local microenvironment. Using combination Northern blot analyses, Taqman RT-PCR, situ hybridizations, endocan was shown to be preferentially endothelial lining tumor xenografts, including human non-small lung cancer, rat glioma, renal carcinoma. In contrast, mRNA at low levels embryos E4.5 E18.5. Under vitro conditions, umbilical vein cells (HUVEC) upregulated cell-conditioned medium, an effect inhibited addition neutralizing antibody growth factor (VEGF). Moreover, treatment HUVEC with VEGF resulted dose- time-dependent increase mRNA. The results suggest vivo its tumor-derived factors.
