摘要: Protoplasts of Nicotiana glauca and N. langsdorffi were prepared from leaf tissue by enzymatic digestion fused with the aid polyethylene glycol. The mixed population protoplasts was grown first on an enriched medium (M3 Kao, et al.) then transferred to a lacking phytohormones, which selects against parental types. A total 174 calli that grew hormoneless obtained. Mature flowering plants differentiated 19 different calli, more than one three these, making 23 regenerated in all. Each shown be parasexual hybrid it formed tumors; corolla, leaf, plant habit similar to, but somewhat from, amphiploid produced cross pollination. hybrids examined cytologically and, instead number 42, they found have range 56 64 chromosomes, accounted for characteristics observed. At meiosis mostly bivalents pollen fertility high, averaging 84 percent. Seeds or progeny been obtained 16 hybrids. unusual chromosome numbers may due primarilymore » triple fusions (giving 60-66 chromosomes) followed losses during callus growth, accompanied selection particular aneuploidy (2n = 64) favorable development plantlets calli.« less
