Selection of T-DNA-tagged male and female gametophytic mutants by segregation distortion in Arabidopsis

摘要: As a strategy for the identification of T-DNA-tagged gametophytic mutants, we have used T-DNA inser- tional mutagenesis based on screening distorted segregation ratios by antibiotic selection. Screening z1000 transgenic Arabidopsis families led to isolation eight lines showing reproducible z1:1, suggesting that these are putative mutants caused insertion at single locus. Genetic analysis transmission through reciprocal backcrosses with wild type showed severe reductions in genetic male and/or female gametes. Direct evidence mutant phenotypes was investigated DAPI staining mature pollen grains and seed set embryo sac morphology cleared ovules. One line, termed limpet pollen, novel phenotype generative cell failed migrate inward after mitosis I, such or sperm cells remained against wall. Two other lines, andarta tistrya, were defective an early arrest development viable megaspore not initiating nuclear division cycles. These data demonstrate efficacy ratio distortion Arabidopsis.
