摘要: In this report, we describe an in vitro culture system for the generation and propagation of axenic amastigotes from well characterised 1S-CL2D line Leishmania donovani. Fine structure analyses these vitro-grown demonstrated that they possessed morphological features characteristic L. donovani tissue-derived amastigotes. Further, (LdAxAm) were shown to synthesise release a secretory acid phosphatase isoform similar produced by intracellular Such LdAxAm also expressed surface membrane 3'-nucleotidase enzyme activity Moreover, LdAxAm, contrast promastigotes, significant levels amastigote-specific A2 proteins. addition, derived long term cultures Ld had infectivity both human macrophages hamsters vivo. Thus, described herein provides useful tool large quantities uniform populations line. The availability such material should greatly facilitate studies concerning cell molecular biology parasite developmental stage.
