摘要: Bacillus spp. are Gram-positive bacteria that secrete a large number of extracellular proteins industrial relevance. In this report, three hydrolytic enzymes, i.e., alpha-amylase, mannanase and chitinase, were cloned over-expressed in Gram-negative Escherichia coli. We found both the native signal peptides E. coli outer membrane protein, OmpA, could be used to direct secretion recombinant enzymes. The expressed enzymes observed as clearing zones on agar plates or zymograms. Determination enzyme activities different cell compartments suggested ability secreted out into culture medium depends time induction, type molecular mass After overnight most (85-96%) harvested from supernatant. Our results suggest various can recognized by machinery. It seems possible other with similar peptide equally well expression systems. Thus, our finding should able apply for cloning production proteins.
