Heterogeneity of Sonic Hedgehog Response Dynamics and Fate Specification in Single Neural Progenitors

摘要: During neural tube patterning, a gradient of Sonic hedgehog (Shh) signaling specifies ventral progenitor fates. The cellular response to Shh is processed through genetic regulatory network (GRN) code distinct fate decisions. This process integrates level, duration and other inputs affected by noise in cell position. How reliably single cell9s profile predicts its choice unclear. Here we use live imaging track progenitors that carry both reporters zebrafish embryos. We found there significant heterogeneity between cells. quantitatively modeled reporter intensities obtain levels over time systematically determined their correlation with multiple models specification. Our input-output analysis shows while no metric perfectly choices, the maximal level correlates best overall choices across anterior-posterior axis.
