摘要: Single-molecule force spectroscopy has provided tremendous insights into the mechanical characteristics of biomacromolecules including proteins, nucleic acids, and sugars. This review provides instrumentation framework for single-molecule using atomic microscopy experimental procedures determining nanomechanics biomacromolecules. The characteristic parameters determined by proteins (unfolding forces, intrinsic unfolding/refolding rates, transition state distances, binding affinities), acids (elastic modulus, persistence length, overstretching percentages, plateau levels), sugars (force spectra) from past 20 years are tabulated their applications discussed.
