Tudor domain proteins in protozoan parasites and characterization of Plasmodium falciparum tudor staphylococcal nuclease.

摘要: RNA-binding proteins play key roles in post-transcriptional regulation of gene expression. In eukaryotic cells, a multitude with several domains/motifs have been described. Here, we show the existence two Tudor domain containing proteins, survival motor neuron (SMN)-like protein and Staphylococcus aureus nuclease homologue referred to as TSN, Plasmodium other protozoan parasites. Activity analysis shows that falciparum TSN (PfTSN) possesses activity is domain. A specific inhibitor micrococcal nucleases, 3',5'-deoxythymidine bisphosphate (pdTp) inhibits well activities protein. PfTSN predominant nuclear localization. Treatment P. pdTp, inhibited vitro growth both chloroquine-sensitive chloroquine-resistant strains falciparum, while four fold concentration pdTp did not any significant effect on mammalian cell line, Huh-7D12. Altogether, these results suggest an essential enzyme parasite's life cycle.
