Preparation and evaluation of ritodrine buccal tablets for rational therapeutic use.

摘要: Ritodrine hydrochloride (RD-HCl) tablets containing alginate (AL) and lactose (LC) with or without microcrystalline cellulose (MC) as excipients were produced a buccal dosage form. The RD-HCl (2 mg) AL/LC but no MC swelled dissolved gradually in the vitro dissolution test. tablet showing fastest highest drug release rate, called Tablet A1, was selected to show rapid prolonged absorption. However, vivo absorption test using rats, it could not give plasma concentration over human minimal effective level (15 ng/mL). modified AL, LC, (4 mg), named B/MC, showed better hardness faster release. B/MC gave within 15 min, maintained at >15 ng/mL 4 h. Moreover, deconvolution analyses demonstrated that high rate be achieved best B/MC. improved pharmacokinetic profile comparison A1 solution LC are suggested possibly useful for treatment of premature labor.
