摘要: Mouse embryonic stem (ES) cell cultures exhibit heterogeneity and recently are discovered to sporadically enter the 2-cell (2C)-embryo state, critical for ES potency. Zscan4 could mark sporadic 2C-state of cells. However, factors that regulate Zscan4(+)/2C state remain be elucidated. We show Tbx3 plays a novel role in regulation state. activates genes including Tcstv1/3, but not vise versa. Ectopic expression results telomere elongation, consistent with lengthening. Mechanistically, decreases Dnmt3b increases Tet2 protein levels, reduces binding subtelomeres, resulting reduced DNA methylation derepression at e.g. Zscan4. These data suggest can activate by negative repeated sequences, linking maintenance self-renewal
