Electrophoretic separation of acidic and basic proteins in the presence of micromolar concentrations of an ionic liquid

摘要: We report on the use of an ionic-liquid (IL) as additive to acidic background electrolyte (BGE) for capillary electrophoretic separation proteins human serum albumin, transferrin, trypsin inhibitor, hemoglobin, catalase, myoglobin and lysozyme. The method was carried out in a zone electrophoresis mode because concentration ionic liquid is far below its critical micelle concentration, were detected by measurement capacitively coupled contactless conductivity. addition IL BGE micromolar improves (a) detection sensitivity method, (b) recovery protein due alleviated wall-adsorption proteins, (c) resolution strong interaction between protein. In fact, even microheterogeneous variants hemoglobin catalase could be resolved. Proteins separated within typically 18 min at pH 2.5, with limits 0.7 9.4 μg mL−1 (34.8–141.1 nM). specifically employed analyze samples.
