摘要: Cell surface proteins play an important role in multidrug resistance (MDR). However, the identification involving chemoresistant features for cell is a challenge. To identify potential membrane markers hematologic cancer MDR, we used cell- and antibody-based strategy of subtractive immunization coupled with comparative screening leukemia lines from sensitive HL60 resistant HL60/DOX cells. Fifty one antibodies that recognized expressed differently between two were generated. One them, McAb-5D12 not only recognizes its antigen but also block function. Comparative analysis immunofluorescence, flow cytometry, mass spectrum validated nucleoprotein-polypyrimidine tract binding protein associated splicing factor, PSF. Our results identified PSF overexpressed on cells compared relocation nuclear to was common hematological malignancy marrow patients. Furthermore, found contributed sensitivity by inhibiting proliferation. The represent novel potentially useful biomarker MDR prediction. enables correlation expression levels functions some cell-drug response traits using antibodies.
