Germline promoter hypermethylation of tumor suppressor genes in gastric cancer

摘要: AIM: To explore germline hypermethylation of the tumor suppressor genes MLH1, CDH1 and P16INK4a in suspected cases hereditary gastric cancer (GC). METHODS: A group 140 Chinese GC patients whom primary had developed before age 60 or who a familial history were screened for genes. Genomic DNA was extracted from peripheral blood leukocytes modified by sodium bisulfite. The treated then subjected to bisulfite sequencing specific region MLH1 promoter. methylation status assayed using methylation-specific PCR. Clonal allelic positive samples performed obtain comprehensive analysis CpG island these promoter regions. RESULTS: Methylation gene detected only 1/140 (0.7%) patient group. However, this pattern mosaic rather than which has previously been reported non-polyposis colorectal (HNPCC) patients. We found that 10% alleles methylated, consistent with 20% cells having one methylated allele. No detected. CONCLUSION: Mosaic epimutation is present China but at very low level. Germline not frequent event.
