摘要: Cancer growth and metastasis depends on the availability of energy. Energy-sensing systems are critical in maintaining a balance between energy supply utilization for tumor growth. A central regulator this process is AMP-activated protein kinase (AMPK). In times deficit, AMPK allosterically modified by binding increased levels AMP ADP, making it target specific kinases (AMPKKs). signaling prompts cells to produce at expense motility, opposing actions insulin factors. Increasing activity may thus prevent proliferation cells. Activated also suppresses aromatase, which lowers estrogen formation prevents breast cancer Biguanides can be used activate AMPK, but many different interacting factors; understanding these factors important order control abnormal processes that lead neoplasia. Fatty acids, estrogens, androgens, adipokines, another sensor, sirtuin-1, alter phosphorylation activation AMPK. Isoforms differ among tissues serve functions. Targeting regulatory points other than upstream AMPKKs provide additional approaches prevention neoplasia, growth, metastasis.
