摘要: This chapter describes the development of a high-resolution, multimode digital imaging system based on wide-field epifluorescent and transmitted light microscope, cooled charge-coupled device (CCD) camera. The three main parts this are Nikon FXA Hamamatsu C4880 CCD camera, MetaMorph system. presents various design criteria for instrument major features microscope components—the camera upright can produce high resolution images both illumination without switching objective or moving specimen. functional aspects set-up be considered in terms optics, epi-illumination transillumination focus control, vibration isolation table. is somewhat specialized microtubule mitosis studies, it also applicable to variety problems cellular imaging, including tracking proteins fused green fluorescent protein live cells. valuable correlating assembly dynamics individual cytoplasmic microtubules (labeled by conjugating X-rhodamine tubulin) with membranes endoplasmic reticulum DiOC6) cell cortex (by differential interference contrast) migrating vertebrate epithelial plays an important role analysis mitotic mutants powerful yeast genetic Saccharomyces cerevisiae.
