Molecular cloning and evolutionary analysis of the calcium-modulated contractile protein, centrin, in green algae and land plants

摘要: Centrin (= caltractin) is a ubiquitous, cytoskeletal protein which member of the EF-hand superfamily calcium-binding proteins. A centrin-coding cDNA was isolated and characterized from prasinophyte green alga Scherffelia dubia. PCR amplification primers were used to isolate partial, homologous sequences algae Tetraselmis striata Spermatozopsis similis. Annealing analyses suggested that centrin single-copy-coding region in T. S. similis other studied. Centrin-coding regions dubia, encode four colinear domains putatively bind calcium. Phylogenetic analyses, including Chlamydomonas reinhardtii land plant Atriplex nummularia, demonstrate centrins are congruent arose two-fold duplication an ancestral EF hand with Domains 1+3 2+4 clustering. The also those calmodulins demonstrating that, like calmodulin, ancient within ancestor all eukaryotes via gene duplication. relationships inferred comparisons mirror results small subunit ribosomal RNA sequence suggesting useful evolutionary markers algae.
