摘要: The discrimination between perfect-match and single-base-pair-mismatched nucleic acid duplexes was investigated by using oligonucleotide DNA microarrays nonequilibrium dissociation rates (melting profiles). RNA versions of two synthetic targets corresponding to the 16S rRNA sequences Staphylococcus epidermidis (38 nucleotides) Nitrosomonas eutropha (39 were hybridized probes (18-mer 19-mer) a set having all possible single-base-pair mismatches. melting profiles probe-target determined in parallel an imposed temperature step gradient. We derived optimum wash for each probe target simple formula calculate index This corresponded output independent analysis customized neural network program. These results together provide experimental analytical framework optimizing mismatch among on microarray.
