Aberration of epidermal growth factor receptor expression in bone and soft-tissue tumors: protein overexpression, gene amplification and activation of downstream molecules.

摘要: In order to evaluate the involvement of epidermal growth factor receptor, and analyze correlation between gene aberration protein expression in mesenchymal tumors, we examined by immunohistochemistry 125 cases bone soft-tissue tumors. Furthermore, amplification receptor was determined fluorescence situ hybridization. Positive immunostaining found 23 (18.4%). Among these cases, one malignant fibrous histiocytoma showed highest degree (3+) overexpression as clusters hybridization signals, indicating homogeneously staining regions. The second case also a higher (2+) coamplification with centromeric regions, polysomy chromosome 7. levels observed were confirmed immunoblotting be comparable. Moreover, although phosphorylated detected those two histiocytoma, constitutive activation extracellular signal-related kinase 1/2 not observed, suggesting that does necessarily constantly lead signal transduction downstream molecules. remaining 123 including 21 exhibiting weak (1+) immunoreactivity, no nor found. Collectively, infrequently but 'overexpression' is rare can attributed an increase copy number, resulting from or polysomy.
