Stearoyl-CoA desaturase 1 gene expression is necessary for fructose-mediated induction of lipogenic gene expression by sterol regulatory element-binding protein-1c-dependent and -independent mechanisms.

摘要: Stearoyl-CoA desaturase (SCD) synthesizes oleate necessary for the biosynthesis of triglycerides and other lipids. Mice with a targeted disruption SCD1 gene are deficient in tissue have reduced expression sterol regulatory element-binding protein (SREBP) its target genes. The SREBP-1c isoform is known mediator insulin action on hepatic expression, but transcriptional effects due to glucose or fructose still unclear. We found that compared stronger inducer lipogenic causing dramatic increase triglyceride levels. However, when fed SCD1-/- mice, failed induce SREBP-1 genes levels were not increased. Instead feeding caused decrease glycogen plasma induction as well liver triglycerides, glycogen, was partially restored diet supplemented very high (20% by weight) palmitate, stearate, linoleate. Fructose long term induced SREBP-1c-/- further these occurred oleate. Our observations demonstrated produced SCD fructose-mediated through SREBP-1c-dependent -independent mechanisms suggested important lipid carbohydrate homeostasis.
