摘要: Abstract The precise mechanism by which insulin elicits its effects remains to be fully determined. A glycophospholipid, isolated from H35 cells, has been proposed as a possible precursor for an insulin-generated second messenger that mediates the intracellular of insulin. This glycolipid contains hexosamine moiety, inositol, galactose and palmitate. We have cultured rat hepatocytes exhibits chromatographic radiolabelling characteristics similar this precursor. can radiolabelled with glucosamine, galactosamine palmitate, but not myristate or myo-inotisol. Incorporation radiolabel into was insensitive presence either (10−7 M) phosphatidylinositol-specific phospholipase C (PI-PLC) in culture medium. used exhibited normal responses respect glycogen turnover gene expression. Treatment partially purified PI-PLC nitrous acid did result generation aqueous soluble phosphooligosaccharide indicating cleaved agent. is contrast reported cleavage glycolipids found hepatoma lymphocytes. These results question role putative mediator transduction system activated
