摘要: Mechanical stress is a key pathogenic driver of apoptosis in the tubular epithelium obstructive nephropathy. Heat shock protein 70 (Hsp70) and Wilms’ tumor (WT-1) have been proposed to represent linked downstream effectors cytoprotective properties NO. In present study, we sought evaluate whether effects l-arginine neonatal nephropathy may be associated with NO-dependent increases WT-1 Hsp70 expression. Neonatal Wistar–Kyoto rats were submitted complete unilateral ureteral obstruction (UUO) treated thereafter vehicle, L-NAME or by daily gavage for 14 days block augment NO levels, respectively. Normal rat kidney epithelial cells NRK-52E exposed mechanical vitro presence absence L-NAME, l-arginine, sodium nitroprusside (SNP), l-arginine + SNP l-arginine/L-NAME. Induction mRNA expression genes assessed. decreased following UUO coincident increased apoptosis. treatment reduced restored levels control levels. basal apoptotic rates prevented response strain, an effect enhanced SNP co-incubation. anti-apoptotic action l-arginine. experimental reduces restoration directly inhibits strain-induced manner vitro. This potentially implicates NO-Hsp70-WT-1 axis
