摘要: This study examined the relationship between Mycobacterium tuberculosis global clades and patient origin. In UK, rate of is higher amongst patients who originated from Indian subcontinent (ISC), where two dominant lineages are present, Central Asian Strain (CAS) East African (EAI) lineage. Mycobacterial interspersed units containing variable number tandem repeats DNA fingerprinting M. strains isolated UK ISC, as defined by novel software, identified that CAS was most prevalent clade (39%) EAI third (15%). To further elucidate host origin strain lineage, rigorous new models infection were developed which assessed mycobacterial growth cell response. The monocyte-derived macrophage model more appropriate for measuring cytototoxicity than THP-1 in absence infection, 50% cells died compared to 2% macrophages. caused 1.5 fold necrosis their four model. Finally, response polarised macrophages Caucasian donors different compared. grew preferentially better M2 donors. prevalence Midlands likely be due a combination specific importation increased ability this transmit population present Midlands.
