摘要: Publisher Summary This chapter presents the procedure for assaying and purification of second component human complement—C2. The assay C2 is based on hemolytic reaction degree lysis measured from A410 supernatant. 100% hemolysis 0% tubes are included. Results expressed in 50% units multiplied by number erythrocytes per ml. In purificathm outdated citrated plasma made 20 mM CaCl 2 left to clot overnight at 4°C. then centrifuged down serum stored – 20°C. steps include: euglobulin precipitathm;;chromatography CM-Sephadex C-50; ammonium sulfate precipitation; chromatography aged CNBr-activated sepharose-4B; diethylaminoethyl -sepharose; sepharose-4B-bound anti-factor B antibody. fraction represents a about 4000-fold yield 8 mg liters serum. homogeneous criteria sodium dodecyl -polyacrylamide electrophoresis N-terminal analysis.
